The development of OMT's fully human monoclonal antibody platform had three major milestones. The generation of: (1) light chain transgenic founder animals, (2) heavy chain transgenic animals, and (3) knockout rats with inactivated immunoglobulin loci.
OMT has created knockout and transgenic rats to serve as founders. At the end of 2008, OMT generated knockout rats with mutated immunoglobulin heavy chain loci (KO F0). Science featured this milestone in “Knockout Rats Produced via Embryo Microinjection of Designed Zinc Finger Nucleases” (2009, 325:433). In 2010, the European Journal of Immunology described the knockout rat in “Characterization of Immunoglobulin Heavy Chain Knockout Rats” (2010, 40:2932-2941).
